合成物
由于分子的特性肯定來源于其合成物,氨基酸分析結果見表1。一些觀點的重點在于:大量的親水殘基產生高度帶電,高度可溶的分子;天冬氨酸與谷氨酸的相對豐度,特別是與精氨酸和賴氨酸比較時,賦予分子強酸性質;半胱胺酰殘基普遍存在,其中涉及到二硫鍵和色氨酸缺失。這種缺失的影響之一是人體白蛋白對紫外線的低吸收率(見表2)。比較來看,人體免疫球蛋白G的色氨酸與絡氨酸含量表示為蛋白質中每千個總殘基中特定氨基酸的殘基 。白蛋白吸收系數低得多的事實主要反映了色氨酸水平的差異。

The abundance of albumin in circulating human plasma appears to be matched only, by the abundance of albumin papers in the biochemical 1iterature. The present review has therefore been written, not in an attempt to survey the information exhaustively, but with the intent of illustrating various viewpoints from which chemists have examined albumin. I’m attempting to synthesize the kaleidoscopic pictures emerging from these examinations,?some preference has been given to the properties that seem most likely to be important when albumin is used clinically. Little attention has been devoted to genetic, phylo genetic, or immunological aspects of the molecule, except as they concern its chemical properties. Furthermore, even though human albumin has been isolated from such diverse sources as platelets (1), thyroid gland (2), and urine (3), the work reviewed is that carried out with albumin obtained from placental (4) or venous (5) plasma.
COMPOSITION
Since the properties of a molecule must stem from its composition, the results of amino acid analysis are shown in Table 1. Several points bear emphasis:the large number of hydrophilic residues, producing a highly charged, highly soluble molecule; the relative abundance of aspartic and glutamic acid, especially as compared with arginine and lysine, giving the molecule a strongly acid character; the prevalence of cysteinyl residues,all but one of which are involved in disulfide bridges?and the paucity of tryptophan.
Among the effects of this paucity is the low ultraviolet absorption exhibited by human albumin (Table 2)。 For comparison, the tryptophan and tyrosine content of human immunoglobulin G, expressed as residues of the particular amino acid per thousand total residues in the protein is shown. The fact that albumin has a much? lower absorption coefficient is primarily a reflection of the differ ence in tryptophan levels.
